Just cant get my head around it

If an item costs £300 per 1000 and the resale value is £280 per 1000, what is the percentage difference between the 2.

Im thinking that 300 per 1000 = 100% so there is a difference of 20 (300 -280)

which would now be 20 per 1000 difference,

Does this then work out 20/1000 * 100 = 2%

Is this correct ?????

Almost. It is the 300 (and not the 1000) which is the 100% so the answer is
20/300 * 100 = (20/3)% ~ 6.67%
