I have an equation "-r2=-1+P0/P". I have to get r2 on its own. Can I change the minus and plus signs as follows, "r2=1-P0/P"?

Looks good to me! (But, technically, let's call it "multiplying through by -1", okay?) ;)

Whadda heck is Po ?
If standard (for algebra, at least) conventions are being used, \(\displaystyle P_0\) will be the "initial" value (say, the value at time t = 0) for whatever is represented by \(\displaystyle P\). ;)
